I suggest using HopStop.com for help getting around New York via public transit - they'll tell you when subways aren't running on the normal routes (which happens often) and reroute you more efficiently than Google Maps.
Take the NJ Transit from the train station connected to the airport - there are multiple lines that work, but you want one that goes to New York Penn Station. Exit from Penn Station at W 32nd St and 7th Ave. Walk east on W 32nd St towards 6th Ave - there will be an entrance near W 32nd St and Broadway for the QNRW subway line. Take one of these trains to Canal/Broadway. That's round Little Italy.
